Separation and Purification of Antimicrobial Substance from Syzygium aromaticum Merrill et Perry for Treatment of Microbial Vaginosis

Abstract

|
Syzygium aromaticum Merrill et Perry (SA) has traditionally been used as a preventive agent and medicine against labor pain and infection. This study was conducted to isolate the antimicrobial substance from SA that is effective against microbial vaginosis. While hot water and ethyl acetate extracts from SA did not show any antimicrobial activity, methanol extract at 50 mg/mL showed antimicrobial activity against Chryseobacterium gleum KCTC 2904, Proteus mirabilis KCTC 2510, Sphingomonas paucimobilis KCTC 2834, and Candida albicans ATCC 10231. Among 95 fractions obtained by silica gel column chromatography, fraction No. 23 had the greatest antimicrobial activity and broad antimicrobial spectrum against C. gleum KCTC 2904, P. mirabilis KCTC 2510, S. paucimobilis KCTC 2834, and C. albicans ATCC 10231. Moreover, the Rf 0.85 spot produced by TLC of fraction No. 23 showed antibacterial activity against C. gleum KCTC 2904. Isolation and purification of the single peak at a retention time (Rt) of 24.989 min was achieved by HPLC of Rf 0.85 spot, and the peak at Rt 24.989 min showed antibacterial activity against C. gleum KCTC 2904, P. mirabilis KCTC 2510, S. paucimobilis KCTC 2834, and C. albicans ATCC 10231.
